Principle:
The below enzymatic sequence is employed in this amylase kinetic assay:

Reagents Supplied: 60 tests (10 x 16ml)(Concentrations refer to reconstituted reagents):
- Glucoamylase (Rhizopus Sp.) 10,000IU/L.
- p-Nitrophenyl D-Maltoheptaoside 0.9mM.
- Glucosidase (yeast) 25,000IU/L.
- Calcium Chloride 5mM.
- Sodium Chloride 50mM.
- Buffer 50mM, pH 6.9.

Performance Characteristics:
1. Linearity: 1,500 IU/L
2. Sensitivity: Based on an instrument resolution of 0.001 absorbance, the present amylase kinetic assay procedure has a sensitivity of 5 IU/L.

Expected Values:
Urine: 18 to 330 IU/L.
Serum: up to 96 IU/L.

Interfering Substances:
1. Macroamylasemia increases the pancreatic amylase in serum.
2. Young et al. has listed certain drugs and other substances that are known to interfere with amylase.
3. A number of diseases affect the determination of amylase.
4. Some bacteria, Insulin, Lipemia, hemolysis, and lipemia will also increase amylase activity.
