The below enzymatic sequence is employed in this AST, SGOT liquid reagent assay:

Reagents Supplied:  150 tests (5 x 25ml + 5 x 5ml )
AST Liquid Reagent 1 and 2 are clear, colourless liquids. After combining AST Liquid R1 (Buffer Reagent) and AST Liquid R2 (Co-Enzyme) the NADH 0.18mmol/L Stabilizers and Preservatives reagent contains:
- LDH (rabbit muscle) > 600 U/L
- NADH 0.18mmol/L
- L-Aspartate 240mmol/L
- Tris Buffer, pH 7.5 80mmol/L
- 2 – Oxoglutarate 12mmol/L
- MDH (porcine muscle) > 600 U/L
- Stabilizers and Preservatives

Performance Characteristics:
1. Comparison: A group of 125 sera ranging in AST activity from 13 – 399 U/L was assayed by the described AST, SGOT liquid reagent method and by a similar commercially available AST reagent. Comparison of the results yielded a correlation coefficient of 0.999 and the regression equation was y = 0.964x + 0.964. (Comparison studies were performed as specified by NCCLS Tentative Guideline, EP9-T.)
2. Linearity: AST liquid reagent is linear to 500 U/L at 37°C.6 Performed according to NCCLS Guideline EP6-P.
3. Sensitivity: Based on an instrument resolution of A = 0.001, the method presented shows a sensitivity of 2.65 U/L.

Expected Values:
Normal Range: obtained using SGOT, AST Liquid Reagent (Kinetic) was between 8 – 33 U/L (37°C)

Interfering Substances:
- Hemolysis must be avoided with this SGOT liquid reagent.
- Bilirubin levels up to 40mg/dL.
- Triglyceride levels up to 2000mg/Dl.
- Certain drugs and other substances.
