Intended Use:
Human 8-OHdG Check ELISA is a method to be used for quantitative analysis of the oxidative DNA adduct 8-hydroxy-2’-Deoxyguanosine (8-OHdG) levels in  saliva, urine, plasma, tissue, serum or any other biological samples.

Procedure:
[1]. Monoclonal antibody, the sample or standard are firstly added to the microtiter 96 well plate. The monoclonal antibody will then react competitively with Human 8-OHdG that is bound on the plate and the 8-OHdG in samples solution. Higher concentration of 8-OHdG in sample solution results in a reduced binding activity of antibody to the 8-OHdG on the plate.
[2]. Antibodies bound to the 8-OHdG in the sample are then washed away from the antibodies that have bound to the 8-OHdG coated on the plate.
[3]. A secondary antibody (enzyme labelled) is then added to the plate, this will bind to the monoclonal antibody that is bound to the 8-OHdG coated on the plate.
[4]. Unbound HRP conjugated secondary antibody is removed by another washing step.
[5]. Addition of the substrate solution causes the development of colour, the amount of colour developed is proportional to the amount of anti 8-OHdG antibody bound to the plate.
[6]. Finally, the reaction is terminated by phosphoric acid, and absorbance at 450 nm is measured.
