Reagents Supplied:

Intended Use:
Alpha MSH EIA Kit (Human, Mouse, Rat)(EK-043-01) is designed for in vitro quantitative determination of alpha Melanocyte Stimulating Hormone (alpha-MSH) levels in any biological fluid, tissue homogenate, culture media, plasma, CSF, urine and serum samples.

Assay Principle:
1. Alpha melanocyte stimulating hormone EIA kit has been pre-coated with secondary antibody and also the non-specific binding sites have been blocked.
2. Secondary antibody is able to bind with the Fc fragment of the primary antibody, the Fab segment is competitively bound by both biotinylated peptides and the peptide standard or the targeted peptide in samples.
3. Biotinylated peptide can then interact with the SA-HRP, which then catalyzes the TMB substrate solution and hydrogen peroxide to form a blue coloured solution.
4. Reaction is stopped by adding hydrogen chloride, which forms a yellow coloured solution.
5. Intensity of the yellow solution is inversely proportional to the amount of the peptide in standard solutions or samples.
6. Unknown concentration in samples can then be determined using this Alpha-MSH EIA kit by extrapolation from a standard curve.

Background:
Melanocyte Stimulating Hormones ( Alpha-MSH) are a group of peptide hormones that are produced by cells in the intermediate lobe of the pituitary gland. It belongs to a group called the melanocortins, which also includes ACTH, beta-MSH, beta MSH and alpha MSH. It is an important melanocortin for pigmentation and it functions to stimulate the production/release of melanin by melanocytes in the skin and hair. MSH is also produced by a subpopulation of neurons in the arcuate nucleus of the hypothalamus. Alpha-MSH released into the brain by these neurons has effects on appetite and sexual arousal.
