Intended Use:
Apelin-12 EIA kit (Human, Rat, Mouse, Bovine)(Cat. No.# EK-057-23)
is intended for quantitative assay of apelin 12 levels in urineany biological fluidserum, plasma, tissue homogenate and CSF.

Assay Principle:
1. Apelin-12 EIA kit has been pre-coated with secondary antibody and also the non-specific binding sites have been blocked.
2. Secondary antibody is able to bind with the Fc fragment of the primary antibody, the Fab segment is competitively bound by both biotinylated peptides and the peptide standard or the targeted peptide in samples.
3. Biotinylated peptide can then interact with the SA-HRP, which then catalyzes the TMB substrate solution and hydrogen peroxide to form a blue coloured solution.
4. Reaction is stopped by adding hydrogen chloride, which forms a yellow coloured solution.
5. Intensity of the yellow solution is inversely proportional to the amount of the peptide in standard solutions or samples.
6. Unknown concentration of aplin 12 present in the samples can then be determined using this apelin-12 EIA assay by extrapolation from a standard curve.

Background:
Apelin 12 is an endogenous ligand for the G-protein-coupled APJ receptor. It is widely expressed in various organs such as the heart, gastrointestinal tract, kidney, lung, adipose tissue, endothelium, brain, adrenal glands and human plasma. Apelin 12 functions as a mediator of the cardiovascular control, which includes blood flow and blood pressure. It is also a potent stimulators of cardiac contractility, playing a vital part in cardiac tissue remodeling. Its levels are increased in left ventricles of patients with chronic heart failure.
